BSI-721 is a novel antibody-drug conjugate targeting cadherin-17 (CDH17) for the treatment of gastrointestinal cancers. It is composed of a fully humanized anti-CDH17 monoclonal antibody (mAb) conjugated to monomethyl auristatin E (MMAE) via a cleavable peptide linker. In a CDH17 high-expressing gastric cancer model (SNU-5), treatment with BSI-721 induced significant tumor regression (achieving partial response) and robust tumor growth inhibition (TGI). Significant TGI was also observed in a pancreatic cancer model (AsPC1) with low-to-moderate CDH17 expression. Collectively, these data demonstrate the excellent therapeutic potential of BSI-721 across gastrointestinal malignancies with varying levels of CDH17 expression.
