BSI-532 is a next-generation tetravalent bispecific antibody targeting IL-7R and IL-4R, thereby achieving a "quadruple blockade" of the IL-7, TSLP, IL-4, and IL-13 signaling pathways. By simultaneously acting on upstream drivers (IL-7 and TSLP) and downstream effectors (IL-4 and IL-13), BSI-532 establishes a higher barrier to resistance and achieves durable remission. In head-to-head comparisons with bempikibart and dupilumab, BSI-532 demonstrated superior in vitro efficacy, exhibiting synergistic inhibition of T-cell proliferation and effectively suppressing both the Th1 and Th2 inflammatory axes simultaneously. Furthermore, it has been engineered to possess a potentially prolonged half-life. BSI-532 is indicated for the treatment of chronic inflammatory diseases driven by T-cell immunity or Type 2 (T2) inflammation, including: severe asthma and COPD (tailored for patients with T2-low or mixed inflammatory phenotypes, extending coverage beyond the standard T2-high population); refractory atopic dermatitis (capturing IL-4R non-responders or inadequate responders by concurrently targeting skin-resident memory T cells); psoriasis; and vitiligo. Currently, the project is advancing through IND-enabling studies at full speed, with the IND filing anticipated in the first half of 2027 (1H 2027).
